FOUR SUBSTANCES, DIFFERENT HEALTH QUESTIONS
What can studies of these four substances tell us?
Peptide drugs and NAD+ products are studied for different possible changes in health. This comparison explains who was tested, the strongest findings and the questions each substance leaves unanswered.
Why does each substance need a different next study?
Peptide drugs can alter the body's work, while NAD+ takes part in cell repair and the use of energy. For you, their tests leave different health questions open, as this comparison explains.
BPC has improved wound healing mainly in animals, rather than in human trials. Your injury needs a fair comparison in people, beyond further demonstrations of healing in rats.
People receiving retatrutide lost weight and had better blood sugar control in trials. Fewer later strokes, heart attacks or deaths would need longer studies counting those events.
Pills supplying ingredients for NAD+ increased blood NAD+, without establishing slower ageing in other tissues. Measuring more people's blood wouldn't establish useful repair inside the brain or muscles.
Bremelanotide has large trials and approval for distressing low desire in women still having periods. The same benefits and risks haven't been established for people outside that group.
A general promise of more research doesn't identify which health question will be tested. Your concern helps determine which missing study could supply the needed answer.
How many people were tested, and for how long?
Medicine approval is absent for BPC-157, and sport prohibits its use. In the small human safety report, two adults received treatment into a vein [1].
Both adults received treatment once [1], while researchers checked health tests and side effects [1]. Large human healing trials and measurements of removal from the human body remained missing [2][3].
Retatrutide is undergoing Phase 3 testing, larger comparisons of benefit and safety. An earlier Phase 2 trial, checking possible benefit, assigned 338 adults to groups by chance [11].
Reports reach 48 weeks of follow-up [10][11], with improvements in weight, sugar tests and liver fat [10][11][12]. Later illness and weight after treatment stops aren't reported in this evidence [8].
NAD+ products are sold as supplements, with NMN's legal position disputed. Tests of ingredients for making NAD+ lasted eight to ten weeks [14][15][17].
The ten-week trial checked how women's muscles responded to insulin [15]. Across the different trials, researchers also checked blood NAD+ and walking distance [14][15][17], with few direct tissue tests supporting ageing claims [13].
The FDA approved PT-141 in 2019 for distressing low desire in a defined group of women. Two Phase 3 comparisons enrolled 1,267 women whose periods had not ended permanently [20].
Safety checks reached 52 weeks without a placebo comparison throughout [21]; the trials measured small changes in distress and desire [20]. Human scans and animal tests left the detailed brain explanation unsettled [18][19].
For you, a large study is useful only alongside knowing what researchers checked. A health concern can remain unanswered even when a carefully conducted trial involved many people.
Why do better rat tendons leave your injury unanswered?
BPC-157 improved healing after rats' Achilles tendons were cut right through [6]. The heel bone and calf muscle are attached by this tendon; stomach ulcers also improved [5], with blood vessel tests suggesting one explanation [4].
The described animal findings showed several kinds of improved healing fairly consistently. No comparable human trial establishes whether treatment improves healing of your injured tendon.
In 2025, reviewers found three small early reports about BPC-157 given to people [2]. The larger human comparisons needed for healing remained absent despite substantial animal research.
Rats and beagle dogs were also tested for entry into blood and removal from the body [3]. Following muscle shots, the share reaching rats' blood was about 14 to 19 percent.
For dogs, about 45 to 51 percent reached blood, roughly three times the rat share. That difference cannot establish how much would reach your blood after treatment.
Within less than 30 minutes, half the substance was gone from the animals' bodies [3]. Human timing hasn't been measured, so animal findings cannot justify schedules discussed online.
Why does weight lost today not settle later heart health?
Retatrutide studies fairly compared drug treatment against placebo, containing none of the drug [11][12]. BPC-157 hasn't had matching human healing tests comparing groups in that way.
During testing, retatrutide reduced weight and liver fat, and improved blood sugar. Your concern may instead be future heart attacks, kidney damage or years of life.
A change in a test doesn't automatically establish fewer illnesses in later years. A review from 2025 describes the longer health studies continuing, with results not yet reported [8].
Published follow-up described here reaches 48 weeks [10][11], much less than several years. Larger tested amounts raised heart rate more, with the highest rate around week 24 [11].
The reports don't establish how heart rate changed once those checks stopped. Nor do they establish whether weight stayed off after treatment ended.
NAD+ pills also have clear blood-test results, without establishing repair inside other tissues. Measuring blood cannot settle what happened inside your brain or muscles.
These substances therefore need different studies to answer the question you have. Retatrutide needs later health results; NAD+ needs useful tissue changes checked directly.
Why do tissue tests and the group of women matter?
NR supplies an ingredient for making NAD+; blood NAD+ rose 22, 51 and 142 percent over eight weeks [17]. NMN supplies another ingredient and raised blood readings over 60 days [14].
Those are clear human blood findings, without establishing slower ageing. Your question about the brain or muscles needs useful changes checked inside those tissues.
The review from 2025 found little human benefit and few direct tests of certain tissues [13]. Additional blood-only tests wouldn't fill the missing answer about useful repair elsewhere.
PT-141 was mainly tested in women before menopause, when periods end permanently. Two Phase 3 trials enrolled 1,267 women [20]; further 52-week testing followed 684 women [21].
Approval covers distressing, generally low desire that developed while women still had periods. The label describes half the drug gone from their bodies after about 2.7 hours [22].
The same benefit and safety results are missing for men and for women past menopause. You cannot assume that one tested group's response establishes another group's response.
Women's written answers showed small improvements in desire and distress that supported approval [20]. Researchers still disagree about whether that benefit is noticeable enough in everyday life.
Approved help with desire doesn't establish help with erections or other sexual performance. Even many participants leave a question unanswered when the people you mean weren't studied.
Which claims need more evidence before you can rely on them?
Claims about these substances often extend beyond the changes that researchers actually measured. Your concern involves the hoped-for benefit as well as the contents of the product.
Faster repair of a rat tendon [6] doesn't establish faster recovery for a person. A scan showing less liver fat [10] doesn't establish repaired liver scarring.
Higher NAD+ in blood [17] doesn't establish slower ageing within the body's tissues. Increased desire in women still having periods [20] doesn't establish the same benefit in everyone.
These limits help you match the claimed benefit with the finding that supports it. A wider health claim needs its own study, beyond an encouraging result measuring something narrower.
Material labelled BPC-157, retatrutide or PT-141 for research lacks approved medicines' official checks. Unknown contents, strength and cleanliness add concerns apart from the studied substance's effects.
A pharmacy-made NAD+ injection was recalled because of harmful material left behind by bacteria. The recall was in the most serious category, showing a danger from the product itself.
Further research needs to check specific unanswered questions, beyond repeating the encouraging findings. BPC-157 needs human removal measurements [3], while retatrutide needs later health results [8].
NAD+ needs direct tissue testing [13], and PT-141 needs studies in other groups [22]. Naming those missing tests helps explain what remains unknown for your health.
